Trump was beginning to speak and was gently roasting his past agent (in the audience) who set up his TV show, the Apprentice (I've never seen it).
Apparently the show was taken over by Schwarzenegger after Trump left and has done very poorly.
He was roasting his good Christian friend (and The Arnold) by telling this joke.
Ever have your pastor tell a joke during a sermon?
Certainly got everyone's attention!

The meme in the OP is taken way out of context - this is an actual transcript of both speeches (forgive the site):

selective editing at best, you could have easily compared Obama's remarks about his motorcade causing traffic, to this excerpt of Trump's speech:

It was the great Thomas Jefferson who said, the God who gave us life, gave us liberty. Jefferson asked, can the liberties of a nation be secure when we have removed a conviction that these liberties are the gift of God. Among those freedoms is the right to worship according to our own beliefs. That is why I will get rid of and totally destroy the Johnson Amendment and allow our representatives of faith to speak freely and without fear of retribution.

and that was really their problem with this speech, not some remarks about the tv show. Trump is egging it on to increase Arnold's ratings, in the hope people turn in for the finale to see what the fuss is all about. Worked with me.
